MS2_MS2-PP7_PP7 RNA
Our bacterial three-hybrid (BH3) construct (inspired by Berry and Hochschild, 2018) is composed of two fusion proteins with MS2 and PP7 coat protein and RNA. This present part is composed of two MS2 (from Escherichia phage MS2) and two PP7 (Pseudomonas phage PP7) stem-loops. MS2 and PP7 coat proteins are able to recognize specifically MS2 and PP7 stem-loops, allowing them to link all parts of the construct. In our project MS2_MS2-PP7_PP7 RNA is used as a positive control for the BH3 system. The sequence between the two MS2 and two PP7 contain 6 differents restrictions sites: PstI, AvaI, XmaI, BsoBI, TspMI, and SmaI. This part permits to create a new RNA sequence bordered by MS2 and PP7 stem-loops.
